Diagnostic utility of the loop-mediated isothermal amplification (LAMP) for detection of cytomegalovirus DNA, Epstein Barr and herpes simplex 1 and 2 in biological samples: a systematic review

Abstract

Introduction

Viral infections cause significant morbidity and mortality in immunocompromised and non-immunocompromised individuals. The completion of an early etiologic diagnosis offers great advantages in the treatment of patients, as appropriate and timely antiviral treatment may improve prognosis. This article demonstrates the diagnostic utility of the Loop-mediated isothermal amplification (LAMP) assay for the detection of deoxyribonucleic acid (DNA) of cytomegalovirus, Epstein Barr and herpes simplex 1 and 2 from biological samples.

Materials and Methods

We developed a rigorous systematic review of scientific literature on the subject during the years 2000 to 2009, using different databases. The inclusion and exclusion criteria defined for the articles were applied independently by two investigators and, finally, three studies were considered for data analysis. resultsThe findings show a simple, rapid method for detecting microorganism by LAMP test when compared with its predecessor PCR assay. In addition, the test has a good performance with a sensitivity of 80% to 86%, specificity of 85.7% to 100%, positive predictive value of 66.7% to 100% and negative predictive value of 90% to 99.4%.

Discussion

According to published studies where the LAMP assay was used, this test is a molecular diagnostic tool similar to real-time PCR in terms of sensitivity and specificity, as demonstrated also in the studies selected for this review. The assay was evaluated using clinical samples and the results indicated the suitability and simplicity of the test as a rapid, field diagnostic tool. There are still very few original studies on the usefulness of the LAMP assay, however, this review presents a contribution to the field and provides clarity on the real and valid applications of the LAMP test.
