Comparison between the use of amsonic acid and calcofluor white for staining fungi on agar blocks

Introduction: Early and accurate diagnosis of invasive fungal in-fections is critical to improving the quality of life of immunocompromised patients and to reduce fatal complications. Although staining procedures that are currently implemented are useful, their low specificity and low positive predictive value diminish their effec-tiveness in diagnosis. This is why it is necessary to ex- plore new staining agents, such as stilbene derivatives that are able to emit fluorescence by joining bonds present in the fungal cell wall.
Objective: To identify the operational characteristics of the stain-ing technique with amsonic acid and compare them with those of Calcoflour White.
Results: Specificity of the microscopy analysis was higher with Amsonic acid than with Calcofluor White (100% and 49.4% respectively). The sensitivity was 96.7% with Calcofluor White and 93.5% with amsonic acid.
Conclusions: Amsonic acid is a more specific technique than Calco-fluor White and both staining procedures require pre-vious training for the microscopic detection of fungi.
