Evaluation of genotoxic potencial of colombian phytoproducts active against Leishmania and Trypanosoma by using of the Ames Test and the Comet Assay

Abstract

Leishmaniasis and Chagas diseases are important causes of morbidity and mortality in Colombia and tropical areas. Treatments currently used show high toxicity and several limitations. Therefore, the search for new drugs is imperative. Meanwhile, the great biodiversity of these regions is an abundant source of potential antiparasitic products. In this study, we evaluated two active fractions extracted from a plant of the genus Sapindus, in the scope to evaluate their possible clinical use. Their mutagenic and genotoxic potential were assessed using the Ames Test and the Comet Assay on human primary peripheral blood cells. The results show that these fractions (3S-E y 3S-F) at three concentrations evaluated: 0.3, 3, and 30 μg/ml are not mutagenic in Salmonella typhimurium, strains TA-100 and TA-98. The Comet Assay with the 3S-E fraction indicated statistical significance between the doses 3 and 30 μg/ml and the negative control (DMSO 1.5%), showing the capacity of this fraction to affect ADN even at doses of 3 μg/ml. Subsequent studies are then required in order to determine the genetic damage produced by 3S-E fraction in human cells at the effective doses. Interestingly, peripheral blood cells treated with the 3S-F fraction presented no genotoxicity at all. The data make suggest that this fraction may be an important candidate in the development of therapeutic products against Leishmaniasis and Chagas disease.
