Abstract

Plants were obtained from chusquines of Guadua angustifolia through in vitro regeneration of axilary buds. The evaluated desinfection methods were: mercury dichloride (HgCl2) and sodium hipoclorite (NaClO). The best culture medium was composed by MS salts and vitamins supplemented with 6 Benzylaminopurine 2,5-5,0 mg/l, myo-Inositol 10 mg/l, sucrose 30 g/l and 2,5 g/l Gelrite®. Buds were tested during four weeks for contamination; the final number of established buds and the influence of the physiological age were evaluated. Somatic embryogenesis was also achieved. Proembryogenic calli from axilary buds was induced with 2,4-D. Calli were transferred to media without auxin where embryogenic structures were observed. The results suggest the presence of a regeneration process through somatic embryogenesis on Guadua angustifolia, which has not been described before.
