MODIFICACION DEL METODO DE FIELD DILUIDO PARA LA COLORACION DE MUESTRAS DE SANGRE EN EL DIAGNOSTICO DEL PALUDISMO
Abstract
The staining of blood has always been complicated by the great variability offered by methylene blue, from which practically all the dyes used in the staining of blood samples are derived.
The thick drop was initially colored by covering a large thick drop with diluted methylene blue, but as there was almost no visibility in the central part, it was decided to defibrinate the blood to facilitate diagnosis.
Pampana's discovery of the dehemoglobinizing property of the isotonic salts of methylene blue contributed to obtaining better staining by preceding them with a pretreatment with a solution of phosphate methylene blue.
By means of this previous step, stains are obtained that offer clean fields for microscopic observation, facilitating the identification of blood elements and parasites.
Field's method of rapid staining for thick drops consists of a 1 to 3 second dip in solution “A” (a mixture of methylene blue and phosphates), followed by a brief rinse in distilled water and a similar dip in solution “B” (a mixture of eosin and phosphates).
